LDHA is a direct target of miR‐30d‐5p and contributes to aggressive progression of gallbladder carcinoma

Abstract: Gallbladder cancer (GBC) is the most general biliary tract malignancy, with poor prognosis due to rapid tumor progression and lack of specific symptoms. Lactate dehydrogenase-A (LDHA) can promote Warburg effect to produce lactate and Adenosine Triphosphate (ATP) in aerobic condition, which contributes to oncogenesis metastasis and drug resistance in various cancers. However, the expression and functional role of LDHA in GBC are largely unknown. We determined that LDHA was over-expressed in GBC tumor tissues co… Show more

“…Western blot analysis was performed as previously reported. 22 Briefly, total cell protein lysates were separated by 10% SDS-PAGE and transferred to a nitrocellulose membrane (Beyotime, China). After incubation with specific antibodies and 5% skim milk powder, the NC membranes were then incubated with specific primary antibodies, followed by HRP-labeled secondary antibody (Beyotime, China) and detected by chemiluminescence.…”

“…MicroRNA (miRNA), a kind of small non‐coding RNA, could inhibit translation or promote degradation of targets via combining to the specific region of 3′‐untranslated region (3′‐UTR) in targeted mRNA . Until now, several miRNAs like miR‐34a, miR‐34c, miR‐369‐3p, miR‐374a, miR‐30a‐5p, miR‐142‐3p, miR‐30d‐5p, miR‐323a‐3p, miR‑199a‑3p, miR‐449a, and miR‐4524a/b have been found to target the mRNA of LDHA . In a recent study on colorectal cancer, miR‐34a, miR‐34c, miR‐369‐3p, miR‐374a, and miR‐4524a/b were established to directly inhibit LDHA .…”
