LC-MS/MS versus TLC plus GC methods: Consistency of glycerolipid and fatty acid profiles in microalgae and higher plant cells and effect of a nitrogen starvation

Abstract: Methods to analyze lipidomes have considerably evolved, more and more based on mass spectrometry technics (LC-MS/MS). However, accurate quantifications using these methods require 13C-labeled standards for each lipid, which is not feasible because of the very large number of molecules. Thus, quantifications rely on standard molecules representative of a whole class of lipids, which might lead to false estimations of some molecular species. Here, we determined and compared glycerolipid distributions from three … Show more

“…To determine the lipid molecules that were present in each extract, 2D thinlayer chromatography with lipid extract from cells grown with or without P i was run as described previously (Abida et al, 2015). Each lipid spot was extracted from the silica, and each lipid molecule was identified by mass spectrometry according to Jouhet et al (2017).…”

“…Extraction and quantification were performed with three biological replicates. Glycerolipids were then analyzed and quantified by HPLC-tandem mass spectrometry, with appropriate standard lipids (Jouhet et al, 2017). In brief, lipid extracts corresponding to 25 nmol of total FAs were dissolved in 100 µL of chloroform:methanol (2:1, v/v) containing 125 pmol of each internal standard.…”

“…N and P are often limiting or co-limiting nutrients, and N is often more limiting than P in marine systems (Falkowski et al, 1998). In all microalgae that have been studied so far, either N or P deficiency was the main environmental factor that triggered TAG accumulation (Daboussi et al, 2014;Abida et al;, Jouhet et al, 2017. When N and/or P become limiting, cell proliferation decreases and TAGs start to accumulate as long as organic carbon remains available ( Fig.…”
