LC-MS/MS Screening, Total Phenolic, Flavonoid and Antioxidant Contents of Crude Extracts from Three Asclepiadaceae Species Growing in Jordan

Abstract: This study aimed to evaluate the antioxidant activity and total phenolic content (TPC) and total flavonoid content (TFC) of crude extracts obtained from three Asclepiadaceae species, namely, Calotropis procera L., Peruglaria tomentosa L., and Pentatropis spiralis (Forsk.) Decne. Both butanol and aq. methanol extracts of the three species showed the highest amount of phenol and flavonoid contents, which exhibited the greatest antioxidant activity in the scavenging of 2,2-diphenyl-2-picrylhydrazyl free radical (… Show more

“…To the best of our knowledge, no previous research has been done on the tested species of Pleurospermum to obtain data regarding its bioactive phytochemicals. Our findings were also in agreement with a recent study in which Al-Dalahmeh et al, reported that the highly polar fractions have higher levels of flavonoid and phenolic contents [24].…”

“…The compound of each subfraction was analyzed using the LC-MS/MS method ( Supplementary Materials ). This method has better selectivity, sensitivity, and accuracy for fast analysis [ 37 , 38 ]. According to LC-MS/MS analysis results, subfractions C, D, E, and F obtained eighteen compounds.…”

“…The biosynthesis of flavonoids, flavones, flavonols, isoflavonoids, and anthocyanins was greatly enriched in the DAMs. Flavonoids, flavones, flavonols, isoflavonoids, and anthocyanins have strong antioxidant activity [ 43 , 44 , 45 ]. These findings might explain why TV, TQ, and TM had some higher levels of TP and TF, as well as better antioxidant activity.…”

“…In the correlation analysis, Acacetin-7-O-glucuronide, 5,7,4′-Trihydroxyisoflavone-7-O-galactoside-rhamnose, 3′,4′,5,6,7-Pentamethoxyflavanone, Wogonin-7-O-Glucuronide, and Apigenin-7-O-(2″-feruloyl) glucuronide showed strongly positive correlations at a level of p ≤ 0.01 with APC. Previous studies have also shown that flavonoids have a strong antioxidant activity [ 43 , 44 , 45 , 46 , 47 ]. For example, Huang et al found Wogon-in-7-O-Glucuronide alleviates colitis by improving the intestinal epithelial barrier function via the MLCK/pMLC2 pathway [ 48 ].…”
