Layering defect in p35 deficiency is linked to improper neuronal-glial interaction in radial migration

Abstract: Several genes essential for neocortical layering have been identified in recent years, but their precise roles in this process remain to be elucidated. Mice deficient in p35--an activator of cyclin-dependent kinase 5 (Cdk5)--are characterized by a neocortex that has inverted layering. To decipher the physiological mechanisms that underlie this defect, we compared time-lapse recordings between p35(-/-) and wild-type cortical slices. In the p35(-/-) neocortex, the classic modes of radial migration--somal translo… Show more

“…A similar branched phenotype was reported in migrating neurons in the cortex of the p35 mutant 66 , where it was thought to contribute to altered cortical lamination. Since p35/CDK5 complex interacts with DCX, it is possible that this interaction relates to the branched phenotype 67 .…”

“…p35 is the most important regulatory subunit of Cdk5 during neurodevelopment, [38][39][40] and we confirmed that it interacts with Cdk5 during cell death. 28 Cleavage of p35 to p25 by calpain has been linked to activation of Cdk5.…”

“…In contrast, radially migrating neurons seem to have a single leading process (Rakic 1972). Interestingly, defects in the interaction between pyramidal cells and radial glial fibers frequently lead to aberrant branching of the leading process (Elias et al 2007;Gupta et al 2003), which suggest that the bipolar morphology of radially migrating neurons might be imposed by glialguided locomotion.…”

“…Ndel1 is required for targeting Lis1 and dynein to the centrosome, and facilitates the nucleation and anchoring of microtubules to the centrosome (Guo et al 2006). In addition, Ndel1 facilitates the interaction between Lis1 and dynein and regulates dynein-mediated retrograde transport (Gupta et al 2003;Li et al 2005;Mesngon et al 2006;Smith et al 2000). Disruption of Lis1 function abolishes centrosomenucleus coupling, increasing the distance between the two organelles.…”
