Lateral resolution enhancement of confocal microscopy based on structured detection method with spatial light modulator

Abstract: Lateral resolution in confocal microscope is limited by the size of pinhole. In this paper, we attempt to introduce a new method to achieve structured detection through using spatial light modulator (SLM) to improve it. SLM modulates the Airy disk amplitude distribution according to the detection function in collection arm. Instead of using CCD to capture spot images and modulate them with numerical analysis in virtual structured detection (VSD), this method uses SLM to accomplish these aims with higher imagin… Show more

“…For example, scanning structured illumination microscopy 126 increases the spatial resolution of laser scanning microscopy with patterned illumination or detection. [127][128][129][130][131][132] Image scanning microscopy [133][134][135] increases the resolution of confocal microscopy [136][137][138] by a factor of ffiffi ffi 2 p simply by replacing the point detector of the confocal microscope with an array detector. Moreover, these super-resolution scanning microscopy methods provide optical sectioning, thereby enabling imaging of relatively thick biological samples.…”

“…SIM has been widely used in optical sectioning, phase imaging, and 3D surface measurement [7]. As an expanded case, digital grating was introduced into virtual structured detection [8,9,10], for modulating measured patterns.…”

“…Among the investigations of lateral calibrations, many studies focus on the optical system [12]. For example, H. Ni et al proposed a new method to achieve structured detection using a spatial light modulator, which modulates the Airy disk amplitude distribution according to the detection function in the collection arm [13] and B. Wang et al presented confocal microscopy with structured detection in a coherent imaging process to achieve a higher resolution with a comparably large pinhole [14], however, the systematic geometric errors which adversely affect the relative position and orientation between measuring probes and measurands are usually neglected [15,16]. B. Daemi et al designed a comprehensive verification test by using a high precision metrology method based on subpixel resolution image analysis [17].…”
