Latent transforming growth factor β1 activation in situ: quantitative and functional evidence after low‐dose γ‐irradiation 1

Abstract: The biological activity of transforming growth factor beta1 (TGF-beta) is controlled by its secretion as a latent complex in which it is noncovalently associated with latency-associated peptide (LAP). Activation is the extracellular process in which TGF-beta is released from LAP, and is considered to be a primary regulatory control. We recently reported rapid and persistent changes in TGF-beta immunoreactivity in conjunction with extracellular matrix remodeling in gamma-irradiated mouse mammary gland. Our hypo… Show more

“…Detection of apoptosis by TUNEL analysis was performed using the Apoptag Detection Kit (Serologicals Corp., Norcross, GA, USA) according to the manufacturer's instructions. Immunofluorescence for Smad2 and active TGFb1 was performed using fresh frozen tissue cryosections as described previously (Ehrhart et al, 1997) using the following antibodies: active TGFb1 (cat # AF-101-NA, lot FS08, R&D Systems, Minneapolis, MN, USA), Smad2/3 (Santa Cruz Biotechnology, Santa Cruz, CA, USA), and secondary antibodies labeled with Alexa 488 or 594 fluorochromes (Molecular Probes, Eugene, OR, USA). Images were captured using a scientific-grade 12-bit charge-coupled device (KAF-1400, 1317 Â 1035 6.8-mm 2 pixels) digital camera (Xillix, Vancouver, Canada).…”

“…Barcellos-Hoff and colleagues 39 have shown that irradiation of the mammarygland stromal component promotes the tumorigenic potential of non-irradiated epithelial cells. These investigators had previously shown that even low levels of irradiation lead to remodelling of the ECM in breast tissue and activation of latent transforming growth factor-β (TGF-β), which affects tissue and organ function 40 . Moinfar et al 41 examined genetic alterations in tumour-associated stroma from several independent cases of mammary carcinoma, and found chromosomal rearrangements that were not present in the malignant carcinoma cells.…”

“…Tais resultados indicam ativação "in situ" da proteína TGFβ1 em células do parênquima pulmonar. Em outros tecidos, como a glândula mamária, observou-se ativação da TGFβ em horas e em dias após irradiação 19,33 . Sugeriu-se que a ativação precoce seria conseqüente ao "stress" oxidativo induzido pelos radicais livres produzidos pela radiação e a elevação continuada e tardia da proteína devido aos mecanismos celulares via plasmina.…”
