Laser Desorption/Ionization Mass Spectrometric Imaging of Endogenous Lipids from Rat Brain Tissue Implanted with Silver Nanoparticles

Abstract: Mass spectrometry imaging (MSI) of tissue implanted with silver nanoparticulate (AgNP) matrix generates reproducible imaging of lipids in rodent models of disease and injury. Gas-phase production and acceleration of size-selected 8 nm AgNP is followed by controlled ion beam rastering and soft landing implantation of 500 eV AgNP into tissue. Focused 337 nm laser desorption produces high quality images for most lipid classes in rat brain tissue (in positive mode: galactoceramides, diacylglycerols, ceramides, pho… Show more

“…Other ToF‐based systems include the spiralToF (Jeol, Peabody, MA, USA) that allows high mass resolution imaging (Muller et al . ) as well as the MALDI ion trap–ToF (Shimadzu Axima, Kyoto, Japan), which facilitates MSn fragmentation and enhanced structural characterization, respectively (Shimma et al . ).…”

“…Recent reports have demonstrated the use of SL for preparation of well‐defined model catalysts through deposition of mass‐selected clusters, nanoparticles, and organometallics . SL also has been used to deliver nanoparticles to tissue where they serve as an ionization enhancing matrix for mass spectrometry imaging . Other applications of SL include purification and conformational enrichment of peptides and proteins, chiral amplification of molecules, and preparation of thin films .…”

“…Schnapp et al described a silver foil blotting technique for the detection of many classes of olefins . Most other researchers have cited the silver sputtering approach but instead introduced different flavors of silver nanoparticles (AgNPs) sprayed onto samples as an IMS approach for detecting olefins and other endogenous species such as beta‐amyloid peptides, phospholipids, and TAGs . While nanoparticles are exciting and provide the sensitivity and spatial resolution required in IMS, they can be especially difficult to prepare for those without the proper training and know‐how to reproduce, even when the synthesis recipe is followed to the dot.…”
