LAPTM5 Plays a Key Role in the Diagnosis and Prognosis of Testicular Germ Cell Tumors

Abstract: Objective. Testicular germ cell tumors (TGCT) are a serious malignant tumor with low early diagnosis rates and high mortality. Methods. To investigate novel biomarkers to predict the diagnosis and prognosis of this cancer, bioinformatics analysis was used as an accurate, efficient, and economical method. Results. Our study detected 39 upregulated and 589 downregulated differentially expressed genes (DEGs) using the GEO and TCGA databases. To identify the function of DEGs, GO functional analysis, three pathway … Show more

“… 23 Besides, LAPTM5 mRNA expression was elevated in testicular germ cell tumors, and its high expression contributed to poor OS. 9 LAPTM5 also served as a biomarker for patients with lung cancer, exhibiting the correlation between LAPTM5 and methylation. 24 Additionally, LAPTM5 mRNA expression was decreased in neuroblastoma cell lines and primary tumors by DNA methylation around its transcriptional start site.…”

“… 8 E3 protein encoded by LAPTM5 might represent an essential role in hematopoiesis. 9 Additionally, LAPTM5 is preferentially expressed in immune cells and hematopoietic cells, and it has a close interaction with Nedd4 family of ubiquitin ligases. LAPTM5 is an active regulator of the proinflammatory signaling pathways in macrophages.…”

“…As a highly malignant tumor, there is an urgent need to find effective diagnostic and prognostic targets for identifying early-stage patients, developing proper treatments, and improving ACC's poor prognosis. Therefore, using bioinformatics techniques to unravel the genomic properties of ACC at the molecular level is crucial for finding effective treatments and predicting patient survival and relapse risk, and there have been several successful cases of bioinformatics used in cancer research [48][49][50][51].…”

“…Cell proliferation-related hallmarks, such as the terms "mitotic spindle", "G2/M checkpoint", and "DNA repair", and immunerelated hallmarks, such as the terms "interferon gamma response" and "inflammatory response", were enriched. In previous literature, the studied biomarkers have been shown with prognostic impact in various malignancies (29)(30)(31). Among them, the role of GLB1L and TMEM265 is consistent with our study, but the mechanism is unknown.…”
