Lapatinib Plus Local Radiation Therapy for Brain Metastases From HER-2 Positive Breast Cancer Patients and Role of Trastuzumab: A Systematic Review and Meta-Analysis

Abstract: Background: Intracranial activity of lapatinib has been demonstrated in several studies in patients with human epidermal growth factor receptor-2 positive breast cancers (HER-2+ BC). Stereotactic radiosurgery (SRS) has been increasingly used as the local therapy for brain metastases in breast cancer patients. Increased objective response rate was observed for lapatinib plus whole brain radiotherapy (WBRT) is such patients with high toxicity. Objective: We seek to obtain clinical evidence of synergistic efficac… Show more

“…In this series, the role of lapatinib in association with radiation treatment was mainly investigated. Lapatinib given concurrently with brain stereotactic radiotherapy showed not only a decreased local failure rate (5.7% vs. 15.1%, p < 0.01) but was also associated with a reduced rate of RN (1.3% vs. 6.3%, p < 0.01) compared to SRT alone [ 28 , 29 ].…”

“…As mentioned above, glycogen metabolism plays an important role in the cancer progression [ 32 ]. Several lines of evidence suggest that the different isoforms of PYGs are over-expressed and involved in the maintenance [ 33 ], progression and invasiveness [ 34 , 35 ] of different cancer cells.…”

“… 41 Bicalutamide is used to treat metastatic hormone-sensitive prostate cancer. 42 Lapatinib is widely used in patients with non-small cell lung cancer, 43 breast cancer, 44 aggressive prolactinomas 45 and oesophageal cancers. 46 The nonconventional drugs predicted by the study may have a new use in choriocarcinoma.…”
