Language delay aggregates in toddler siblings of children with autism spectrum disorder

Abstract: BackgroundLanguage delay is extremely common in children with autism spectrum disorder (ASD), yet it is unclear whether measurable variation in early language is associated with genetic liability for ASD. Assessment of language development in unaffected siblings of children with ASD can inform whether decreased early language ability aggregates with inherited risk for ASD and serves as an ASD endophenotype.MethodsWe implemented two approaches: (1) a meta-analysis of studies comparing language delay, a categori… Show more

“…A goal in improving the functional outcomes of children with autism is to identify those at greatest risk as early in life as possible, often before the behavioral repertoire of the infant is sufficiently mature to reveal consistent signs of the disorder. Infant siblings of children with ASD have an increased incidence of ASD diagnosis, currently estimated to be as high as 1 in 5 [Ozonoff et al, ], and are also at increased risk for language delay [Marrus et al, ]. Earlier identification of potential language delay in toddlers at risk for ASD would allow for earlier intervention and possibly improve outcomes.…”

“…Proband-sibling associations were further investigated among domains of language development, a reported endophenotype of ASD [37]. Expressive and receptive languages were significantly correlated among sibling pairs, driven by pairs concordant for ASD ( Table 3, Fig.…”

“…With regard to demographic characteristics, gender and educational level of the mother will be taken into account (Marrus et al . , Rescorla , Tager‐Flusberg , Weismer and Kover ). In addition, non‐verbal abilities and ASD features (social–communicative difficulties and repetitive and restricted behaviours) were included as characteristics of the child's development (Marrus et al .…”
