Laminin is the ECM niche for trophoblast stem cells

Abstract: The niche is a specialized microenvironment for tissue stem cells in vivo. It has long been emphasized that niche ECM molecules act on tissue stem cells to regulate their behavior, but the molecular entities of these interactions remain to be fully elucidated. Here, we report that laminin forms the in vivo ECM niche for trophoblast stem cells (TSCs), the tissue stem cells of the placenta. TSCs expressed fibronectin-binding, vitronectin-binding, and laminin-binding integrins, whereas the integrin ligands presen… Show more

“…We isolated ICMs for explant culture by immunosurgical removal of trophectoderm ( Solter and Knowles, 1975 ). ICMs were maintained intact and plated on dishes coated with laminin 111-E8 ( Kiyozumi et al., 2020 ) in N2B27 with or without PD+A83. In N2B27, a central mass of relatively undifferentiated cells persisted in most cases, but patches of trophectoderm morphology often outgrew.…”

“…In particular, laminin LAMA1 was highly expressed in the first trimester, whereas LAMB3, LAMA2, LAMC3 were upregulated in the second trimester. Laminins have an important role during implantation ( Haouzi et al, 2011 ), maintenance of trophoblast stemness ( Kiyozumi et al, 2020 ) and EVT migration ( Zhang et al, 2018b ).…”

“…Nevertheless, laminins mainly dictate the structure and assembly of MB [77]. Moreover, these glycoproteins play a crucial role in cell migration, adhesion, differentiation, and wound healing [78][79][80][81]. They also participate in early embryonic development and organogenesis.…”
