Laminar Analysis of Human Neocortical Interictal Spike Generation and Propagation: Current Source Density and Multiunit Analysis In Vivo

Abstract: Multicontact microelectrodes were chronically implanted in epilepsy patients undergoing subdural grid implantation for seizure localization. Current source density and multiple unit activity of interictal spikes (IISs) were sampled every approximately 150 microm in a line traversing all layers of a cortical column. Our data suggest that interictal epileptiform events in humans are initiated by large postsynaptic depolarizations, consistent with the hypothesis that human IISs correspond to animal paroxysmal dep… Show more

“…The delay for the sink to rise from middle to upper cortical layers was as long as ~500 ms in the current study, whereas in human interictal spikes it is ~20ms (Ulbert et al, 2004). Comparable delays have been found in animal sensory areas (Barth and Di, 1990;Schroeder et al, 1998) and human V2 (personal observation).…”

“…These studies are typically done on rat or cat in in vitro preparations. However, laminar-electrode recordings have been done in awake macaques (Schroeder et al 1998) and even humans (Halgren et al 2006;Ulbert et al 2001Ulbert et al , 2004Wang et al 2005). Thus LPA, combined with template-fitting analysis, offers opportunities for deeper insights into the functional synaptic connections even for these systems.…”

“…Several early studies using microelectrodes indeed showed increased multi - (Altafullah et al, 1986;Ulbert, Heit, et al, 2004), and single unit activity (Babb and Crandall, 1976;IsokawaAkesson et al, 1989;Wyler et al, 1982) during IID generation. Other studies however, found no or limited correlation (Babb et al, 1973;Rayport and Waller, 1967;Thomas et al, 1955;Wyler et al, 1982).…”
