LAMC2 acts as a novel therapeutic target of cetuximab in laryngeal cancer

Abstract: This study was set out to determine the function of LAMC2 in laryngeal cancer (LC). Initially, we identified the expression of LAMC2 in LC cells and tissues using TCGA datasets, GEO datasets (GSE143224), qRT-PCR, and western blot. Besides, we analyzed the correlations between LAMC2 and clinicopathologic features in LC patients. The CCK-8 assays were performed to detect cell viability and the half-maximal inhibitory concentration of cetuximab (IC50) in LC cells. We explored the correlations between LAMC2 and EG… Show more

“…LAMC2 was found to promote the growth of LSCC cells by CCK-8, colony formation, and flow cytometric assays, which was similar to the findings of Wang et al, who found that LAMC2 promoted the proliferation and cell viability of laryngeal cancer cells [ 36 ]. Moreover, LAMC2 promoted LSCC cell migration and invasion.…”

“…In this study, through systematic screening analyses, we report that LAMC2 is an EGFR TKI sensitive gene with both oncogenic and protein transport functions in lung cancer. As a subunit of laminin-332, which is responsible for normal epithelial cell adhesion, differentiation, and migration, LAMC2 is frequently overexpressed in various cancers including, lung cancer [12,13,32,33], laryngeal cancer [34], gastric cancer [35], cholangiocarcinoma [25], thyroid carcinoma [11], and pancreatic ductal adenocarcinoma [36]. In accordance with previous reports, our results indicated that LAMC2 silencing inhibited lung cancer cell progression through reduced cell activity, increased apoptosis, arrest in G1 to S cell cycle progression, and diminished tumor growth in vivo.…”
