Lactogenic Hormone and Cell Type-Specific Control of the Whey Acidic Protein Gene Promoter in Transfected Mouse Cells

Abstract: The whey acidic protein (WAP) is a major milk protein. It is abundantly expressed in mammary epithelial cells, and its gene is controlled by lactogenic hormones. The identification of regulatory cis-acting sequences of the mouse WAP gene was so far dependent on the analysis of transgenic animals. We report here the possibility of analyzing regulatory sequences by gene transfer experiments using the lactogenic hormone-dependent mammary epithelial cell line HC11. A WAP-chloramphenicol acetyltransferase construct… Show more

“…The WAP gene is expressed in mammary epithelial cells during late pregnancy and throughout lactation. Its expression is regulated by a variety of l a c t o g e n i c h o r m o n e s , s u c h a s i n s u l i n , glucocorticoid, and prolactin [11][12][13]. WAPs have a signal peptide [14] and two domain structures with a four-disulfide core (4-DSC) in a conserved arrangement [15].…”

“…Mobility shift and exonuclease protection assays have determined the presences of binding sites for Stat 5 (involved in the prolactin response), glucocorticoid receptor (GR) complex and other factors necessary for transcription in the proximal promoter region (Watson et al, 1991;Welte et al, 1993;Lubon and Hennighausen, 1987;. Deletional analysis of the promoter in transfected HC11 cells has determined two regions between -2500 to -1500 and -450 to -1 that contain hormone response elements (Doppler et al, 1991). Further deletional analysis of the -219 to -1 segment identified an essential hormone induced enhancer region at -195 and a repressor region between -219 and -195 (Mink et al, 1992).…”
