Lactoferricin B like peptide triggers mitochondrial disruption‐mediated apoptosis by inhibiting respiration under nitric oxide accumulation inCandida albicans

Abstract: Nitric oxide (NO) is a potentially powerful weapon against Candida albicans, and the regulation of intracellular NO levels is therefore important for controlling its physiological functions. Lactoferricin B like peptide (LBLP) is a 23‐mer antimicrobial peptide (AMP) derived from the Scolopendra subspinipes mutilans. We confirmed that LBLP treatment led to the generation of endogenous NO in C. albicans, which was associated with the NO synthase pathway. Here, we examined the antifungal activity of LBLP with foc… Show more

“…The cells exposed to diazepam exhibited the main phenotypic characteristics of a process similar to apoptosis, such as exposure to phosphatidylserine and increased defragmentation of the DNA depending on concentration [51,55]. In yeasts, the main sources of DNA damage involve oxygen metabolism and generation of ROSs [56].…”

“…Finally, calcium‐free Krebs buffer was used to wash mixture three times. The fluorescence intensities of Fura‐2AM (excitation = 335 nm, emission = 580 nm) were observed with a spectrofluorophotometer (Shimadzu RF‐5301PC, Shimadzu, Kyoto, Japan) 13 …”

“…The exogenous pathway is mediated by death receptors on the cell membrane; the death ligand binds to death receptors, and activates apoptosis through the death domain, which is not dependent on mitochondrial destruction. 31,32 Mitochondria are important organelles in the process of apoptosis, and a decrease in mitochondrial transmembrane potential is an important feature of early apoptosis. JC-1 is an ideal fluorescent probe widely used in the detection of MMP.…”
