Lactobacillus plantarum YS2 (yak yogurt Lactobacillus) exhibited an activity to attenuate activated carbon-induced constipation in male Kunming mice

Abstract: The present study investigated the effects of Lactobacillus plantarum YS2 (LP-YS2) that was isolated from yak yogurt on activated carbon-induced constipation in Kunming (KM) mice. The KM mice were orally administered LP-YS2 and reference strain Lactobacillus delbrueckii ssp. bulgaricus. Administration of LP-YS2 [1.0 × 10 9 cfu/kg of body weight (BW)] promoted gastrointestinal peristalsis and reduced the first black stool defecation time (129 min), which clearly defines attenuation of the voiding difficulty in … Show more

“…Neurotransmitter also plays an important role in gastrointestinal hormones [28]. A couple of neurotransmitters have been identified to be associated with the motor activity of the gastrointestinal tract, such as Ach, NO, SP, and VIP, which play important roles in the regulation of gastrointestinal motility [14,24,29]. Among them, Ach and SP are excitatory neurotransmitters that promote bowel motility and speed defecation, while NO and VIP are inhibitory neurotransmitters that relax the gastrointestinal tract and inhibit bowel movement [3,30].…”

“…The PCR was performed in an automatic thermocycler (QuantStudio 6 Flex PCR, Life Technologies) with the following parameters: 95°C for 60 s; 40 cycles of 94°C for 30 s, 58°C for 30 s, and 72°C for 50 s; 75°C for 10 min. GAPDH was selected as an internal reference, and the relative expression of cluster of differentiation 117 (c-Kit), stem cell factor (SCF) glial cell-derived neurotrophic factor (GDNF), inducible nitric oxide synthase (iNOS), transient receptor potential vanilloid-1 (TRPV1) and cyclooxygenase-2 (COX-2) mRNA was calculated using the 2 −ΔΔCt formula (Zhao et al, 2019). Note: Values presented are the mean ± standard deviation (n = 10/group).…”

“…Constipation is a clinical manifestation of digestive system disorders and is characterized by the formation of hard feces that are difficult to excrete. The prevalence of constipation ranges from 2% to 30%, especially in children and geriatric populations (Chen, Chao, & Pan, 2020;Zhao et al, 2019). Constipation can be divided into primary and secondary constipation, with primary constipation including three types (i.e., normal-transit constipation, slow-transit constipation, and defecatory disorders) (Lembo & Camilleri, 2003).…”

“…Mice in the LC‐YBJ02‐L, LC‐YBJ02‐M, and LC‐YBJ02‐H groups were given 0.2 mL different concentrations of bacterial suspension at 16:00 a day by gavage. The concentrations were 10 8 colony forming units (CFU)/kg, 10 9 CFU/kg, and 10 10 CFU/kg LC‐YBJ02 (Zhao et al., ), and the test lasted for 6 weeks. During the experiment, the weights of the mice prior to gavage were recorded.…”
