Lactate preconditioning promotes a HIF-1α-mediated metabolic shift from OXPHOS to glycolysis in normal human diploid fibroblasts

Abstract: Recent evidence has emerged that cancer cells can use various metabolites as fuel sources. Restricting cultured cancer cells to sole metabolite fuel sources can promote metabolic changes leading to enhanced glycolysis or mitochondrial OXPHOS. However, the effect of metabolite-restriction on non-transformed cells remains largely unexplored. Here we examined the effect of restricting media fuel sources, including glucose, pyruvate or lactate, on the metabolic state of cultured human dermal fibroblasts. Fibroblas… Show more

“…In addition, lactate levels are relatively high in the fetus (~0.5-2 mM in umbilical venous blood at 17-21 weeks gestation [32]) due to high glycolytic activity of the placenta [33], which decreases rapidly after birth (~0.5-1 mM in umbilical venous blood at term delivery [32]) after achieving independence from placental circulation, with maternal arterial lactate levels of 0.68 ± 0.07 mM at delivery [34]. Lactate can also stabilize HIF-1α in fibroblasts [35] and cancer cells Metabolic sensors and mitochondria. Metabolic sensors detect changes in oxygen, nutrient, or hormone levels to induce or inhibit mitochondrial biogenesis or removal by mitophagy.…”

“…Lymphomas are generally considered glycolytic [ 32 ]. As reprogramming of energy metabolism is a hallmark of cancer progression [ 33 ], we questioned whether a shift to oxidative phosphorylation (OXPHOS) occurred in the drug-resistant clone.…”

“…Like differentiated tumor cells, CSC can get their energy from glucose [ 16 , 42 , 43 , 44 , 45 ]. Adoption of a glycolytic metabolism can be driven by microenvironmental constraints, such as a hypoxic environment, or induced by mitochondria dysfunctions [ 46 , 47 , 48 , 49 ]. For example, mutations in genes encoding subunits of the respiratory chain complexes have been associated with the transformation of differentiated tumor cells into CSC [ 50 ].…”
