Lack of ethnic differences of moxifloxacin and metabolite pharmacokinetics in East Asian men

Abstract: This study was designed to investigate ethnic differences in the pharmacokinetics (PKs) of moxifloxacin and its metabolites, M1 (sulfo conjugate) and M2 (acyl-glucuronate), among Japanese, Chinese, and Korean populations, following oral administration. We used a population PK modeling approach using data from a clinical study involving 79 healthy male volunteers. A comprehensive population PK model considering the PK mechanism of moxifloxacin and its metabolites was newly built. The structures of the final mod… Show more

“…10,47 However, we did find that MRP2 transports the moxifloxacin-glucuronide (M2). Based on this, we hypothesized, in agreement with Kaneko et al, 45 that not moxifloxacin itself, but moxifloxacin-glucuronide is transported from the liver to the intestines. This is followed by rapid hydrolysis by gut bacteria, resulting in the excretion of moxifloxacin in the feces, 45,46 similar to bilirubin deconjugation.…”

“…22 Moxifloxacin is mainly metabolized to glucuronide and sulfate conjugates. Exposure to the metabolites was shown to be lower, 52 while apparent half-lives were similar to that of moxifloxacin, 53 which makes the metabolites an unlikely cause of such delay in returning to baseline.…”

“…We searched the reports on race difference of PK parameters of quinolones. Results showed that the PK of levofloxacin, moxifloxacin and ciprofloxacin do not show race difference (Fish & Chow, 1997;North et al, 1998;Hasunuma et al, 2016;Kaneko et al, 2018;Tolentino-Hernandez et al, 2020). For each drug, the PK parameters in various races get close to each other after correction by weight (Fish & Chow, 1997;Kaneko et al, 2018;Tolentino-Hernandez et al, 2020).…”
