Lacisediminihabitans profunda gen. nov., sp. nov., a member of the family Microbacteriaceae isolated from freshwater sediment

Abstract: A novel Gram-stain-positive bacterial strain, CHu50b-6-2T, was isolated from a 67-cm-long sediment core collected from the Daechung Reservoir at a water depth of 17 m, Daejeon, Republic of Korea. The cells of strain CHu50b-6-2T were aerobic non-motile and formed yellow colonies on R2A agar. The phylogenetic analysis based on 16S rRNA gene sequencing indicated that the strain formed a separate lineage within the family Microbacteriaceae, exhibiting 98.0%, 97.7% and 97.6% 16S rRNA gene sequence similarities to G… Show more

“…Amplification of the 16S rRNA gene was done with the PCR method and applying the universal primer set 27F/1492R [14]. The PCR amplification conditions were set up as follows: 95 °C for 5 min, 30 cycles of 95 °C for 1.5 min, 55 °C for 1 min and 72 °C for 1 min and final extension for 10 min at 72 °C [15]. To get the full 16S rRNA gene, sequencing was carried out with following primers: 27F (5′-AGAGTTTGATCCTGGCTCAG-3′), 785F (5′-GGATTAGATACCCTGGTA-3′), 800R (5′-TACCAGGGTATCTAATCC-3′) and 1492R (5′-GGTTACCTTGTTACGACTT-3′) [14, 16].…”

“…The antibiotic susceptibility of strain BN140002 T was checked on R2A agar plates using the paper disc method. The discs contained following antibiotics (μg per disc): nalidixic acid (30), tetracycline (30), amikacin (30), ampicillin/ sulbactam (20), kanamycin (30), vancomycin (30), chloramphenicol (30), teicoplanin (30), streptomycin (25), gentamicin (30), spectinomycin (25), rifampicin (30), lincomycin (15) and erythromycin (30). Other physiological and enzyme activities were assayed using API 20E, API ZYM and API 50CH strips (bioMérieux) according to the manufacturer's instructions.…”

“…For analysis of cellular fatty acids, strain GW1-11 T and the reference strains were grown in TSA broth at 28 ℃ and cells were harvested in the late exponential phase (33). The fatty acids were prepared and identification was carried out according to the standard protocol of the Sherlock Microbial Identification System (34). Polar lipids and quinones were investigated from freeze-dried cells (35), and the polar lipids were identified by two-dimensional TLC as described.…”

“…At the time of writing, there are 59 validly published genera in the family Microbacteriaceae with Microbacterium as the type genus (https://lpsn.dsmz.de/family/chitinophagaceae). Members of the family Microbacteriaceae are Gram-stain-positive, predominantly aerobic, motile or non-motile, non-spore-forming, and have anteiso-C 15 : 0 , iso-C 16 : 0 and anteiso-C 17 : 0 as the major fatty acids [1–6]. During a study on the bacterial diversity in a sludge collected from a pesticide plant (located in Wenzhou, Zhejiang Province, PR China), a small, yellow-pigmented and smooth colony was screened and purified.…”
