Labeling of adipose-derived stem cells with quantum dots provides stable and long-term fluorescent signal for ex vivo cell tracking

Abstract: Stem cells derived from adipose tissue (ADSC) have been used in cell therapy as an alternative to treat chronic and degenerative diseases. Using biomedical and image trials to track the cells when infused in the target tissue is essential to control cell migration and adhesion. The objective of the present study was to label and assess the adhesion of goat adipose tissue-derived stem cells (g-ADSC) after cell infusion in animal models by tracking luminescent intracytoplasmatic nanocrystals. The cells were labe… Show more

“…The variations in gene expression between the ADSC and STZ groups are indicated by arrows, and fold-change values greater than 2 are shown injury, hepatic lipid deposition, glomerular sclerosis and other lesions in rats [54], but ADSCs are widely applied because they are abundant, easily obtained and autologous in origin [55][56][57]. ADSC injection via the tail vein to rodents ① reduced serum ALT and AST levels and mortality [10] and ② increased hepatic Akt and ERK1/2 phosphorylation and serum albumin levels [11], suggesting that ADSCs have a therapeutic effect on acute liver failure and can promote liver regeneration.…”

“…Mesenchymal Stem Cell therapy is an innovative alternative of cell therapy proposed by several studies in the regenerative medicine area, for the treatment of various human and animal diseases, chronicdegenerative or not. This proposal is based on: the in vitro characteristics of low immunogenicity, since they do not express significant levels of the Major Histocompatibility Complex type 2; of immunomodulatory capacity, since they can induce immunosuppression through the production of several chemokines with antiinflammatory properties; in addition to the capacity of plasticity, demonstrated by differentiation in distinct cell lines (Costa et al 2017;Rocha et al 2017;Sironi et al 2017;Xie et al 2018). MSCs are defined by the ability to self-renew and differentiate into specialized cell types.…”

“…In particular, several types of fluorescent nanoprobes have been explored for long-term in situ in vivo fluorescent imaging of tumor. For example, inorganic fluorescent nanoparticles (noble metal nanoparticles, , upconversion nanoparticles, , and inorganic quantum dots , ) have been utilized for optical imaging of cancer at both cellular and animal levels. However, the low biodegradability and potential cytotoxicity limited their applications. In comparison to inorganic fluorescent nanoparticles, organic fluorescent nanoparticles should be an alternative because of their excellent photostability, easy functionality, and good biocompatibility. , Unfortunately, the fluorescent intensity of organic nanoparticles functionalized from many fluorescent molecules would be decreased or quenched in the aggregated state.…”
