Label-free quantitation: A new glycoproteomics approach

Abstract: We demonstrate herein a method for quantifying glycosylation changes on glycoproteins. This novel method uses MS data of characterized glycopeptides to analyze glycosylation profiles, and several quality control tests were done to demonstrate that the method is reproducible, robust, applicable to different types of glycoproteins, and tolerant of instrumental variability during ionization of the analytes. This method is unique in that it is the first label-free quantitative method specifically designed for glyc… Show more

“…One useful strategy for enhancing the MS signal of glycopeptides is to first remove any peptides that may co-elute (25)(26)(27)(28). This strategy has been utilized by multiple groups, taking advantage of a simple enrichment approach using Sepharose beads, as first demonstrated by Wada et al (25).…”

“…In a different approach, the SILAC technique allows incorporation of stable isotope-labeled amino acids into proteins during cell culturing process (196), and was applied to investigate the difference in cell surface N-glycoproteins among different cell types (64). A label-free approach has also been used for glycoproteomics profiling, including a method developed to profile intact glycopeptides in a complex sample (208) and a study that compares the plasma glycoproteome between psoriasis patients and healthy controls (209).…”

“…For alkylated IgA1 HR preparations, cysteine iodoacetamide modification was selected. The relative abundance of each glycopeptide was calculated by use of a label-free quantification method described by Rebecchi et al (37). After assigning all glycopeptide peaks in the spectrum, the ion chromatogram for each glycopeptide ion was individually extracted for the specific glycopeptide ion species Ϯ0.83 m/z.…”
