Label-free oligonucleotide biosensor based on dual-peak long period fiber grating

Abstract: We report the simplification and development of biofunctionalization methodology based on one-step 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC)-mediated reaction. The dual-peak long period grating (dLPG) has been demonstrated its inherent ultrahigh sensitivity to refractive index (RI), achieving 50-fold improvement in RI sensitivity over a standard LPG sensor used in low RI range. With the simple and efficient immobilization of unmodified oligonucleotides on sensor surface, dLPG-based biosensor has been… Show more

“…To active the silicon dioxide of the optical fibers (OFs), which is depicted in the transversal view in Figure 1 A, it was soaked in HCl [5 M] for 30 min, which promotes the generation of hydroxyl groups (see Figure 1 B) on the OF surface, as Zhang and co-workers explain [ 5 ]. Afterwards, the next step was rinsing the OF surface with deionized water and air drying.…”

“…The selective layer can be designed to promote sensors applied to several science fields and highlight biological and biochemical applications [ 2 , 3 ]. In this regard, the state-of-the-art method uses functionalized OF (f-OF) with small molecules, such as organic acids [ 4 ], representing some biomolecules such as DNA [ 5 ].…”

“…Because of the above reasons, changing the reaction path of conjugation to restrain the NSA may be a promising solution. N -Hydroxysulfosuccinimide (sulfo-NHS) can transform the intermediate in a sole EDC process to react with amino groups more efficiently while the sole EDC method retains popularity. Researchers focus on improving coupling efficiency with sulfo-NHS but pay less attention to the advantage of alteration of the reaction path in EDC chemistry. , Moreover, new approaches are required to allow the observation of dynamic events about bioconjugation-induced NSA. This progress should allow the emergence of new findings that will enhance our understanding of the complex molecular interactions between bioconjugates and biofluids.…”
