L-NAME prevents anxiety-like and depression-like behaviour in rats exposed to restraint stress

Abstract: Stressful life events contribute to the development of many neuropsychiatric disorders including depression and anxiety. Animal studies based on the relationship of stress and depression or anxiety are scarce and controversial. Moreover, neither the neurobiological basis of anxiety and depression nor the mechanisms responsible for neurochemical regulation by stressful stimuli are well understood. This study was designed to investigate the possible contribution of both acute (2 h) and chronic (2 h X 15 d) restr… Show more

“…The increase in TNF-α levels is implicated in NF-κB activation and iNOS expression in the brain cortex after immobilization stress (6 h), as used in the present study (4). Inhibition of NOS by a nonselective inhibitor like L-NAME (5,35) or by relatively selective inhibitors of nNOS, like 7-nitroindazole (36,37), produced an antianxiety-like effect. Recently, anxiolytic-like activity of aminoguanidine, an inhibitor of the inducible isoform of NOS (7), was reported in mice from our own laboratory.…”

“…There are several reports that chronic exposure to restraint stress for 15 days (2 hrs daily) leads to the induction of depressive symptoms in rat [32,33]. Chronic injection of corticosterone at an elevated dose has been used in development of stress induced depression model in number of anti depressant studies [34].…”

“…Generally, tail suspension model are used to evaluate behaviour changes as well as immobilization time. Normally depletion of neurotransmitters like serotonin, nor-adrenaline to leads to depression in body 23 . Depression has been linked to perturbations in the neurotransmission involving brain 5-HT, nor-epinephrine (NE) and dopamine activity 27 -28 (Maes and Meltzer, 1995;Posener et al, 1994).…”
