Ku70-deficient embryonic stem cells have increased ionizing radiosensitivity, defective DNA end-binding activity, and inability to support V(D)J recombination

Abstract: V(D)J recombination requires both lymphoid-specific and generally expressed enzymatic activities. All three known generally expressed activities involved in V(D)J recombination are also involved in DNA double-strand break repair (DSBR). Two of these are components of the DNA-dependent protein kinase (DNA-PK) and include Ku80 and DNA-PK catalytic subunit (DNA-PKcs); the third, XRCC4, is a protein of unknown function. The Ku70 protein is an additional component of DNA-PK; Ku70 forms a heterodimer with Ku80 to ge… Show more

“…In addition to error-free NHEJ, evidence from both yeast and mammalian cells strongly supports the notion that Kuindependent NHEJ is an error-prone process (69)(70)(71)(72). BRCA1 may also regulate error-prone end-joining: BRCA1 inactivation was associated with large deletions at the site of the DSB (63).…”

“…In fact, it has been reported that Ku70-deficient mice exhibit decreased levels of Ku86, and vice versa. [25][26][27] We found that 72 h of transfection are sufficient to efficiently knockdown Ku70/86 ( Figure 3e). Quantitative real-time PCR and immunoblotting analyses showed that a significant increase of Apaf1 mRNA and protein levels occurred both in ImimPc-2 and in ETNA cells when compared with cells transfected for 72 h with an unrelated control oligonucleotide (Figure 3d and e).…”

“…As Ku70/Ku80 and DNA‐PKcs modulate RAG‐mediated cleavage6 by assisting the selection of proper RSS combination, this implies that the DNA–PK complex might be involved in the recombination process upstream of RAG cleavage. Indeed, in mice Ku70 and/or Ku80 are absolutely essential for V(D)J recombination and their deletion leads to severe combined immunodeficiency (SCID) 7, 8. As of yet, Ku70/80 mutations have not been identified in patients.…”
