KSHV-Mediated Angiogenesis in Tumor Progression

Abstract: Human herpesvirus 8 (HHV-8), also known as Kaposi’s sarcoma-associated herpesvirus (KSHV), is a malignant human oncovirus belonging to the gamma herpesvirus family. HHV-8 is closely linked to the pathogenesis of Kaposi’s sarcoma (KS) and two other B-cell lymphoproliferative diseases: primary effusion lymphoma (PEL) and a plasmablastic variant of multicentric Castleman’s disease (MCD). KS is an invasive tumor of endothelial cells most commonly found in untreated HIV-AIDS or immuno-compromised individuals. KS tu… Show more

“…Inflammatory cytokine secretion from KSHV-infected cells plays an important role in the pathogenesis of KS and PEL (38). To explore the effects of the drug combination on cytokine secretion, Luminex assays were employed to measure the amount of cytokine secreted into tissue culture supernatants in response to drug treatments.…”

“…Current treatment options for KS include HAART and chemotherapy in the form of liposomal doxorubicin, liposomal daunorubicin, vincristine, bleomycin, etoposide and paclitaxel 14 15. The antiangiogenic drugs, imatinib, sorafenib, bevacizumab and sirolimus, have been used in some cases to counter disease progression and induce regression 16. In cases of single, remote lesions without metastasis, radiotherapy or surgery can prove to be effective forms of treatment 15…”

“…HHV-8 is a DNA virus whose genome consists of a large 165 kb double-stranded DNA molecule and is enveloped by an icosahedral capsid. The virus was originally isolated from KS lesions from an AIDS patient and has been linked to other malignancies besides KS, including primary effusion lymphoma, body cavity-based lymphoma, multicentric Castleman’s disease, germinotropic lymphoproliferative disease, multiple myeloma, angiosarcomas, malignant skin tumors, and squamous cell carcinomas 73. In KS lesions, most of the cells infected by the virus are in the latent phase and do not proliferate or express angiogenic cytokines.…”

“…In KS lesions, most of the cells infected by the virus are in the latent phase and do not proliferate or express angiogenic cytokines. A small percentage remain active and express angiogenic cytokines such as cytokines such as VEGF, IL-6, CXCL8, and bFGF 73. Multiple cell types can be infected by KHSV, but the infection of endothelial cells plays a major role in viral proliferation and angiogenesis.…”
