KRAS Mutation

Abstract: Treatment of colon carcinoma with the anti-epidermal growth factor receptor antibody Cetuximab is reported to be ineffective in KRAS-mutant tumors. Mutation testing techniques have therefore become an urgent concern. We have compared three methods for detecting KRAS mutations in 59 cases of colon carcinoma: 1) high resolution melting, 2) the amplification refractory mutation system using a bifunctional self-probing primer (ARMS/Scorpion, ARMS/S), and 3) direct sequencing. We also evaluated the effects of the m… Show more

“…With optimized sample handling and standardized DNA extraction, it is possible to screen paraffin-embedded samples with higher sensitivity [40], [41]. Despite the increased fragmentation of DNA extracted from paraffin-embedded tissue, it is possible to reliably screen shorter amplification products up to 250 bp in length.…”

“…For first round mutational analysis, DNA samples from selectively microdissected areas were PCR amplified with primers flanking KRAS exon 2 as previously described (35), followed by Sanger DNA sequencing. Positive samples for mutation by Sanger sequencing were not further evaluated, negative samples were evaluated by HRM curve to achieve higher analytical sensitivity.…”

“…Several reports have compared the performance of pyrosequencing, real-time PCR, and Sanger sequencing in detecting KRAS mutation for molecular diagnostics 911. More recently, the Infiniti assay12,13 and 454 next-generation sequencing14,15 limited to discordant cases have been compared with other techniques.…”
