Knockdown of NLRP3 alleviates high glucose or TGFB1-induced EMT in human renal tubular cells

Abstract: Tubular injury is one of the crucial determinants of progressive renal failure in diabetic nephropathy (DN), while epithelial-to-mesenchymal transition (EMT) of tubular cells contributes to the accumulation of matrix protein in the diabetic kidney. Activation of the nucleotide binding and oligomerization domain-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome leads to the maturation of interleukin (IL)-1B and is involved in the pathogenic mechanisms of diabetes. In this study, we explored th… Show more

“…This is confirmed by our previous study that recombinant pyrin domain protein (RPYD) attenuated OVA-induced inflammatory response in asthmatic mice by inhibiting NF-κB signaling (Piao et al, 2019). It is recently reported that knockdown of NLRP3 in human renal tubular cells alleviated TGFβ1-induced EMT (Song et al, 2018). This finding has inspired us to verify whether RPYD could affect the occurrence of EMT in airway remolding asthma mice.…”

“…Studies have shown that TGFβ1 induces and promotes the occurrence of EMT (Haddad et al, 2019;Song et al, 2018). Therefore, TGFβ1 was used as an agonist to induce the occurrence of EMT in BEAS-2B cells.…”

“…cells. Song et al (2018) indicated that knockdown of NLRP3 antagonized HG-induced epithelial-mesenchymal transition by inhibiting ROS production and phosphorylation of SMAD3, P38 MAPK, and ERK1/2. These studies have shown that NLRP3 could regulate the production of ROS, which was consistent with our findings.…”

“…Inflammasomes are involved in the EMT of cancer or other epithelial cells. For example, knockdown of NLRP3 alleviates high glucose or TGF-β1-induced EMT in human renal tubular cells [45]. NLRP3 regulates cellular proliferation and metastasis via EMT and the PTEN/AKT signaling pathway [46].…”
