Knee loading protects against osteonecrosis of the femoral head by enhancing vessel remodeling and bone healing

Abstract: Osteonecrosis of the femoral head is a serious orthopedic problem. Moderate loads with knee loading promote bone formation, but its effects on osteonecrosis have not been investigated. Using a rat model, we examined a hypothesis that knee loading enhances vessel remodeling and bone healing through the modulation of the fate of bone marrow-derived cells. In this study, osteonecrosis was induced by transecting the ligamentum teres followed by a tight ligature around the femoral neck. For knee loading, 5 N loads … Show more

“…Importantly, salubrinal suppressed the maturation of osteoclasts in both early and late development phases, the migration and adhesion of pre-osteoclasts. Furthermore, macrophage (G) and mononuclear (M) were osteoclast progenitors [25]. Salubrinal affected the development of osteoclasts in osteoporosis by CFU-M/CFU-GM assays.…”

“…Bone histomorphometric studies demonstrate that joint loading stimulates bone formation, accelerates fracture healing, and improves bone strength (19)(20)(21). Our previous studies also reveal that joint loading reduces skeletal inflammation and degradation, and it may offer a novel physical regimen for osteoarthritis and osteonecrosis (22,23). Ankle loading, 1 form of joint loading, is an effective means to induce bone formation throughout periosteal and endosteal diaphysis of the tibia by activating signaling pathways, such as PI3K, extracellular matrix-receptor interactions, TGF-b signaling, and Wnt signaling (19)(20)(21).…”
