Klotho Acts as a Tumor Suppressor in Cancers

Abstract: The klotho gene is a classical "aging suppressor" gene. Its roles in the pathology of chronic kidney diseases have been well documented. However, the role of Klotho in tumorigenesis, cancer progression, and prognosis is attracting more and more attention. Recent studies have shown that Klotho participates in the progression of several types of human cancers. Klotho functions as a tumor suppressor by inhibiting insulin/IGF1, p53/p21, and Wnt signaling. Silencing klotho gene expression is mainly mediated through… Show more

“…KLOTHO , a novel cell-cycle regulatory TSG, has been reported to be transcriptionally silenced in MDA-MB-231 cells (Wolf et al, 2008; Xie et al, 2013). Treatment with the combinations of 20 μg/mL GTPs and 10 μM SFN significantly ( P < 0.001) reactivated mRNA expression of KLOTHO by 11.2-, 11.4- and 20.2-fold in MDA-MB-231 cells at 24, 48 and 72 h, respectively, as compared to their respective controls (Fig.…”

“…Expression of p21 CIP1/WAF1 has been shown to be epigenetically silenced in many cancers including breast cancer (Chen et al, 2013a; Chew et al, 2012). In addition to p21 CIP1/WAF1 , KLOTHO, a novel 1012-amino acid transmembrane protein, has also been shown to play major role as a tumor suppressor, mainly by inhibiting IGF-1/insulin pathways in many cancer subtypes (Doi et al, 2011; Lee et al, 2010; Wolf et al, 2008; Xie et al, 2013). In addition, upregulation of KLOTHO has been positively correlated with the suppression of cancer metastasis through inhibition of transforming growth factor-β1 signaling (Doi et al, 2011).…”

“…Since that publication, the role of klotho in tumorigenesis, cancer progression, and prognosis has attracted increasing attention. 23 Recent studies have shown that klotho participates in the progression of several types of human cancer, including lung cancer, gastric cancer and pancreatic adenocarcinoma. 23 In the majority of these malignant tumors, klotho acts as a tumor suppressor gene.…”

“…The physiological and pathological function of KL shows it can be used as a regulator of oxidative stress and senescence (3). Moreover, recent studies show that KL is involved in the progression of several types of human cancers, and plays an important role in tumorigenesis, proliferation, survival, autophagy and resistance to antitumor therapies (4). KL is inactivated through promoter hypermethylation and potentially functions as a tumor suppressor gene in human cancers (5,6), suggesting loss of KL may serve as an early marker for breast tumorigenesis (7).…”
