Kisspeptin/KISS1R System in Breast Cancer

Abstract: Kisspeptins (KP), peptide products of the kisspeptin-1 (KISS1) gene are the endogenous ligands for a G protein-coupled receptor (GPCR) - KP receptor (KISS1R). KISS1R couples to the Gαq/11 signaling pathway. KISS1 is a metastasis suppressor gene and the KP/KISS1R signaling has anti-metastatic and tumor-suppressant effects in numerous human cancers. On the other hand, recent studies indicate that KP/KISS1R pathway plays detrimental roles in breast cancer. In this review, we summarize recent developments in the u… Show more

“…Activation of this G-coupled protein receptor by its canonical ligand, kisspeptin (metastin), has been shown to inhibit tumor cell migration and invasion in several cancers including RCC [48, 49]. Recently, a pro-invasive role has been proposed for KISS1R in breast cancer [50]. Data generated in this study additionally supports an invasive role for KISS1R in ccRCC.…”

“…In vitro and in vivo downregulation of KISS1 in triple-negative breast cancer cells can also take part in the IL30-driven prometastatic program, although contrasting data on the metastasis suppression function of KISS1-derived peptides/KISS1R axis have been reported in breast cancer (39).…”

“…KISS1 encodes for Kisspeptin, which is a peptide ligand for G protein-coupled receptor KISS1R. KISS1 exhibits anti-metastatic effects in numerous human cancers, including melanoma, thyroid, ovarian, bladder, gastric, esophageal, pancreatic, lung, and pituitary cancers [83] [84]. Clinically, reduced KISS1 expression is associated with poor prognosis in cancer patients.…”
