Kinetics of the Solution-Mediated Polymorphic Transformation of the Novel l-Carnitine Orotate Polymorph, Form-II

Abstract: Research studies related to the polymorphs of l-Carnitine orotate (CO), a medication used for the treatment and prevention of liver diseases, are insignificant or almost nonexistent. Accordingly, in the present study, l-Carnitine orotate (CO) was prepared for investigating CO polymorphs. Here, a reactive crystallization was induced by reacting 1g of l-Carn (1 equivalent) and 0.97 g of OA (1 equivalent) in methanol (MeOH); as a result, CO form-I and CO form-II polymorphs were obtained after 1 h and 16 h of stir… Show more

“…As shown in Fig. 4 , L-Car showed numerous distinctive peaks at precise diffraction angles of 2θ-degrees: specifically at 9.2, 16.2, 18.7, 23.9, 28.2 and 37.9 2θ-degrees, in agreement with the literature [ 34 ]. In contrast to L-Car ( Fig.…”

“…Crystalline Forms I and II are very insoluble at pH 4–7, and the solubility of Form II is lower than that of Form I [ 37 ]. Moreover, the increase in supersaturation could significantly promote the rate of nucleation [ 51 ]. Under non-sink conditions, the ASD underwent crystallization to the metastable Form I.…”

“…l -carnitine (3-hydroxy-4- N -trimethylammonium butyrate) is an endogenous compound that plays a pivotal role in fatty acid metabolism [ 1 ]. l -carnitine supplementation is marketed in the form of acetyl- l -carnitine (analogs of l -carnitine) [ 2 ] and carnitine-orotate ( l -carnitine and an ion complex salt of orotic acid) [ 3 , 4 ]. Commercially, the carnitine-orotate complex, a combination of carnitine-orotate, biphenyl dimethyl dicarboxylate, and other minor supplementary compounds, is also available [ 5 ].…”
