Kinetics of albumin- and alpha-fetoprotein-production during rat liver development

Abstract: Synthesis of most of the plasma proteins is one of the main functions of the hepatocytes. Albumin synthesis is quantitatively the most abundant. In the present study we investigated albumin-and alpha-fetoproteingene-expression, and the function of the secretory apparatus during rat liver development. To this purpose we used the method of radioactive biosynthetic labeling of newly synthesized albumin and alpha-fetoprotein (AFP) to monitor the secretory capacity of endodermal cells derived from ventral foregut r… Show more

“…To determine whether the epithelioid cells in the freshly isolated HSC preparations were quiescent HSCs, we evaluated the cells for coexpression of albumin and quiescent HSC markers (desmin or GFAP), reasoning that albumin is a relatively specific marker for hepatocytes and their immediate precursors (45,46). Flow cytometry demonstrated coexpression of albumin and desmin in over 90% of the freshly isolated HSCs ( Figure 7C).…”

“…26 Briefly, mouse hepatocytes were kept in culture 24 h and successively labeled in methionin-free RPMI medium supplemented with 35 S-methionin (100 mCi/300 ml medium pro well) for 6 h. Part of the cells was further stimulated with murine IL-6 at a concentration of 100 ng/ml. After the labeling, cells were washed three times in PBS whereas the supernatants were collected and diluted in 50% lysis buffer containing 1% sodium dodecyl sulfate (SDS).…”

“…Majority of plasmatic proteins is synthesized in hepatocytes, with albumin representing their largest quantitative part (Elmaouhoub et al 2007). Except for albumin (protein), all plasmatic proteins are glycoproteins.…”
