Kinetics of adenylate metabolism in human and rat myocardium

Abstract: A bstractPathways producing and converting adenosine have hardly been investigated in human heart, contrasting work in other species. We compared the kinetics of enzymes associated with purine degradation and salvage in human and rat heart cytoplasm assaying for adenosine deaminase, nucleoside phosphorylase, xanthine oxidoreductase, AMP deaminase, AMP-and IMP-specific 5'-nucleotidases, adenosine kinase and hypoxanthine guanine phosphoribosyltransferase (HGPRT). Xanthine oxidoreductase was not detectable in hum… Show more

“…In both species ecto-5′-nucleotidase exceeds activity of cytosolic 5′-nucleotidases approximately tenfold. Although total 5′-nucleotidase activity of human myocardium was determined to be roughly half that of rat myocardium, the overall distribution of enzymatic activity is similar with the membrane fraction exceeding the soluble fraction 5.5-fold (Kochan et al 1994;Tavenier et al 1995).…”

“…Traces of ATP in the substrates and enzyme preparations were excluded using the ATP Bioluminescent Assay Kit CLS II (Roche Diagnostics GmbH, Mannheim, Germany; Sirius LuminometerBerthold GmbH, Pforzheim, Germany). AdK and ADA were assayed according to Tavernier et al [39]. MK was assayed according to Zhang et al [40].…”

“…It has been characterised in mammals (rat, pig, calf, and human), birds (chicken, goose) as well as a frog [3], the invertebrate Artemia [4] and three fish species [5]. It is best documented in mammals and birds where it has been described in various tissues [1,6], including human (placenta [7], heart [8], erythrocytes [9] and lymphoblasts). cN‐II has a particularly elevated activity in avian liver and in organs with a high turnover rate or DNA replication including human colon carcinoma [10].…”
