Kinetic Studies of the Reactivity of the Sulfhydryl Groups of Glyceraldehyde-3-Phosphate Dehydrogenase

Abstract: The reaction of sulfhydryl groups of glyceraldehyde-3-phosphate dehydrogenase from rabbit and pig muscles with a large molar excess of 5,5'-dithiobis(2-nitrobenzoate) (Nbs,) shows threephasic pseudo-first-order kinetics. Since the fastest reaction between active cysteine-149 and Nbs, is apparently biphasic, half-of-the-sites reactivity towards Nbs, is suggested. Further sulfhydryl groups become reactive as an effect of conformational changes in the protein molecule after formation of a mixed disulfide on cyste… Show more

“…The time curve for the DTNB reaction shown in Figure 1, as well as in previous work [27,28], appears to describe two populations of SH groups in tubulin: fast and slowly reacting. In principle, one might expect at least 20 different rates for the formation of the mixed disulphides [10], although they may not be resolved [36,37]. Kinetic analysis assuming a two-cysteine population model yielded unsatisfactory residuals despite correlation coefficients > 0.99.…”

“…On the other hand, during the last decades there has been developed the theory of functional integral equations of Stieltjes type. Nevertheless, it turns out that a lot of interesting and important problems which can be formulated inside the theory of Volterra-Stieltjes integral equations are not satisfactory solved by the results obtained up to now [16]. In the theory in question, several types of integral operators, both of linear and nonlinear types are investigated in numerous papers and monographs, we refer [17][18][19].…”

“…Glyceraldehyde-3-phosphate dehydrogenase has been reported to have a reactive cysteine at position 151 (Perham, 1969;Banas, Banas and Wolny, 1976), but the ICAT labelled cysteine was at position 247. This suggests that cysteine 247 is also a reactive cysteine but the impact of its modification is yet to be determined.…”
