Kinetic Properties and Ammonium-dependent Regulation of Cytosolic Isoenzymes of Glutamine Synthetase in Arabidopsis

Abstract: Glutamine synthetase (GS; EC 6.3.1.2) is a key enzyme of nitrogen assimilation, catalyzing the synthesis of glutamine from ammonium and glutamate. In Arabidopsis, cytosolic GS (GS1) was accumulated in roots when plants were excessively supplied with ammonium; however, the GS activity was controlled at a constant level. The discrepancy between the protein content and enzyme activity of GS1 was attributable to the kinetic properties and expression of four distinct isoenzymes encoded by GLN1;1, GLN1;2, GLN1;3 and… Show more

“…The presence of such isoenzymes is considered to be crucial for efficient ammonia assimilation in response to variable internal and external environments (7,22,23). We reported significant differences in heat stability and catalytic properties between maize GS1a and GS1d (7).…”

“…In the late response of Arabidopsis, the GS1 family (GLN1;1, ;2, GLN1;3, GLN1;4, GLN1;5;Ishiyama et al, 2004), excluding GLN1;5, was up-regulated, whereas GS2 was repressed. Furthermore, 14 other genes hypothesized to be linked to this pathway were also differentially expressed (Table 2).…”

“…3 The role of Gln1;2 may also be affected by the form of nitrogen supplied to plants because Gln1;2 expression is enhanced by ammonium supply relative to nitrate as the sole N source. 3,4 functions of Gln1;2 are primarily associated with nitrogen remobilization or with N signaling. A high flux of N to juvenile inflorescences is essential for ensuring that a high number of the initiated primordia develop into fertile siliques with a high number of seeds.…”
