Kinetic Modeling of Saccharomyces cerevisiae Central Carbon Metabolism: Achievements, Limitations, and Opportunities

Abstract: Central carbon metabolism comprises the metabolic pathways in the cell that process nutrients into energy, building blocks and byproducts. To unravel the regulation of this network upon glucose perturbation, several metabolic models have been developed for the microorganism Saccharomyces cerevisiae. These dynamic representations have focused on glycolysis and answered multiple research questions, but no commonly applicable model has been presented. This review systematically evaluates the literature to describ… Show more

“…The abundance of Hsp was therefore substantially lower in the absence of oxygen, in line with the lower transcription of Hsp and lower thermo-tolerance of the anaerobic stationary-phase cultures than of the aerobic cultures observed by Bisschops et al [66]. Considering that industrial-scale processes favour anaerobic environments for practical and financial reasons, the present results provide valuable information for the construction of predictive metabolic models [5,9,[68][69][70][71][72][73].…”

“…The before-mentioned SRE approach represents a proven methodology to provide the necessary ground to set up data-driven models [ 46 , 47 , 48 ]. For the organism under investigation, several studies quantitatively investigated the metabolome and transcriptome during a sudden shift from a glucose limitation to excess [ 26 , 47 , 49 , 50 ].…”

“…Existing yeast glycolysis models studied this pathway in isolation (Smallbone et al , 2013; van Heerden et al , 2014). To transit from pathways in isolation, to pathways embedded in a growing cell, inclusion of physiological variables informative of the bioreactor environment is crucial (Lao-Martil et al , 2022). The required data to implement these variables is now available for S. cerevisiae .…”
