Kinetic characterization and molecular modeling of trehalose-6-phosphate phosphatase from Anopheles gambiae and expressed in Pichia pastoris

Abstract: Trehalose-6-phosphate phosphatase (TPP) is one of the primary enzymes involved in the synthesis of trehalose, the main sugar found in insect hemolymph. In the present study, we report for the first time heterologous expression in Pichia pastoris, characterization and homology modeling of TPP from Anopheles gambiae mosquito. Purified TPP recombinant exhibited a molecular weight of approximately 36 kDa with optimum pH of 8.0, optimum temperature of 38°C, and the K M was 3.19 ± 0.10 mM. Inhibition tests revealed … Show more

“…The K M values of BmiTPPC1 and BmiTPPC2 were close to those of reported TPPs (Fig. 6K, L) (Miao et al ., 2016; Shan et al ., 2016; Pessoa et al ., 2017; Liu et al ., 2017a). Interestingly, in spite of the dominance of BmiTPPB transcript in most of the samples, the K M values of BmiTPPB were an order of magnitude greater than BmiTPPC1 and BmiTPPC2 (Fig.…”

“…[6,[9][10] TPP has been studied in a few insects like Delia antiqua, Anopheles gambie, Bactrocera minax, and Sitodiplosis mosellana. [9,6,[11][12] Three TPP genes from Bactrocera minax express highly in the last instar larvae or early pupal stages. [6] The expression and activity of TPP are elevated in the pre-diapause and diapause stages of Sitodiplosis mosellana.…”
