Actinomycetes are the source of many secondary metabolites, enzymes, and antimicrobials that have various applications. Microorganism proliferation is aided in practice by mediated mutations utilizing UV and monochromatic laser radiation. This study aimed to evaluate the photostimulatory effect of UV and laser light on the lipase, amylase and antimicrobial productivity of three actinomycetes' strains from genera: Micromonospora, Pseudonocardia, and Nocardiopsis. The strains were exposed to UV (300-400 nm) and monochromatic laser (632.8 nm) radiations for 10 and 20 minutes. The influence of radiations on the actinomycetes' growth, pigment production, enzymatic and antimicrobial activity was evaluated after incubation at 28 ±2 • C for 7 days. Two reference bacterial strains Escherichia coli NCMB 11943 and Staphylococcus aureus NCMB 6571, and one clinical yeast strain Candida albicans, were used for antibacterial assessment, compared to five commercial antibiotics. The results showed better radiation response of the selected actinomycetes strains to the monochromatic laser. Both Pseudonocardia and Micromonospora have exhibited significant increase in pigmentation with laser exposure. In contrast, Nocardiopsis has shown reduction in pigment production. The UV and Laser radiations have almost doubled the amylase enzyme activity in Pseudonocardia and Micromonospora strains, while not affecting lipase production. On the other hand, the crude metabolites from the Micromonospora sp. strain exhibited a highly promising inhibitory effect against E. coli (NCMB11943) when compared with five commercially available antibiotics. The laser irradiation for 10 min has almost doubled the inhibitory zone of Micromonospora metabolites against S. aureus (from 0.65 to 1.2 cm) and C. albicans (from 0.7 to 1.3 cm). In conclusion, the results of our research recommend using monochromatic laser radiation to enhance the production of amylase and antimicrobial metabolites from Pseudonocardia and Micromonospora, respectively.

Fars province in southern Iran, with an area of 133000 square kilometers is one of the largest provinces of the country. Daptomycin (DPT) was the first lipopeptide antibiotic available for commercialization. It is active against gram-positive bacteria, including resistant strains. This work aimed to develop and validate a turbidimetric microbiologic assay to determine daptomycin in an injectable form. A 3x3 design was employed, at concentrations of 1, 2 and 4.0 µg/mL. The microorganism test used was Staphylococcus aureus ATCC 6538p, and Antibiotic Medium 3 was used as the culture medium. Method validation demonstrated that the bioassay was linear (r=0.9995), precise (RSD=2.58%), accurate (recovery 100.48± 2.11%), and robust. Degradation kinetics was also performed in an alkaline medium, indicating that daptomycin degradation follows first order kinetics under these conditions. The analyses of degraded solutions showed that daptomycin degradation products do not possess bactericidal activity. The bioassay was compared to HPLC method that was previously developed and no significant difference was found between them (p>0.05). The method proved to be appropriate for daptomycin injection quality control.Uniterms: Daptomycin/determination. Daptomycin injection/quality control. O objetivo deste trabalho foi desenvolver e validar um ensaio microbiológico turbidimétrico para quantificar a daptomicina na forma injetável. Empregou-se delineamento 3x3, nas concentrações de 1,0; 2,0 e 4,0 µg/mL. Como micro-organismo teste foi usado Staphylococcus aureus ATCC 6538p, e Meio para Antibióticos nº 3 foi empregado como meio de cultura. A validação do método demonstrou que o ensaio foi linear (r=0,9995), preciso (RSD=2,55%), exato (recuperação de 100,48 ± 2,11%) e robusto. A cinética de degradação em meio alcalino foi avaliada, indicando que a daptomicina segue cinética de primeira ordem nessa condição. A análise das soluções degradadas mostrou que os produtos de degradação da daptomicina não possuem atividade antimicrobiana. O bioensaio foi comparado com o método por CLAE previamente desenvolvido e não houve diferença significativa entre ambos (p<0,05).
