Killing of Cryptosporidium sporozoites by Lactoferrin

Abstract: Intestinal infection caused by is a major contributor to diarrhea morbidity and mortality in young children around the world. Current treatments for children suffering from cryptosporidiosis are suboptimal. Lactoferrin is a glycoprotein found in breast milk. It has showed bacteriostatic and antimicrobial activity in the intestine. However, the effects of lactoferrin on the intestinal parasite have not been reported. In this study, we investigated the anticryptosporidial activity of human lactoferrin on differe… Show more

“…The authors showed possible inhibition of sporozoite viability and infectivity, but no effect was found on the excystation stage of the oocyst wall. These studies suggested that Lf protein or its peptides are impermeable to the thick wall of the oocyst but can inhibit the viability of free forms of this parasite through internalization via the endocytic pathway (Paredes et al, 2017).…”

“…94 For example, LF has inhibitory activity against different stages of Cryptosporidium and can kill Cryptosporidium cysts as well as treat Cryptosporidium-related gastrointestinal infections. 95 Probiotics are an important group of beneficial gut microorganisms, and LF can regulate their formation, enrichment, and equilibrium. 96 The consumption of probiotics may contribute to the balance of beneficial microbiota in the gastrointestinal tract, and the property of LF to bind iron allows it to promote the growth of bif idobacteria and lactobacilli.…”

“…Some studies have combined LF with probiotics and demonstrated that LF inhibits the growth of particular pathogens, thus influencing the composition of the gut microbiota by inhibiting gut pathogens . Probiotics are an important group of beneficial gut microorganisms, and LF can regulate their formation, enrichment, and equilibrium .…”

“…The gardicidal effect of lactoferrin and lactoferrin-derived peptides was also observed at low concentrations, where they caused dilation of the endoplasmic reticulum (ER) membranes, expansion of the nuclear membrane, and plasma membrane protrusions [ 211 ], although high concentrations cause severe morphological changes or even induce programmed cell death [ 212 , 213 ]. Lactoferrin also exhibited antiparasitic activity against Cryptosporidium parvum sporozoites but had no significant effect on oocysts viability or parasite intracellular development [ 214 ].…”
