KIAA1199 overexpression is associated with abnormal expression of EMT markers and is a novel independent prognostic biomarker for hepatocellular carcinoma

Abstract: PurposeTo determined KIAA1199 expression and investigate its correlation with the clinicopathologic data and prognosis of hepatocellular carcinoma (HCC), as well as markers of epithelial-mesenchymal transition (EMT); N-cadherin, E-cadherin and vimentin.Materials and methodsWestern blot, quantitative real-time PCR, and immunohistochemical staining were used to measure KIAA1199 expression in human HCC specimens. Subsequently, the correlation between KIAA1199 expression and the pathological characteristics of HCC… Show more

“…Tumor progression is a complex process regulated by numerous genes over multiple steps, whereas EMT is a process of self-repair that occurs after damage to adult epidermal cells ( 6 , 19 ). During tumor cell development, EMT causes the loss of epithelial characteristics and gain of mesenchymal cell traits ( 22 , 23 ), bestowing tumor cells with stronger invasion and migration capabilities. Downregulation of E-cadherin, which is associated with cellular adhesion, is considered the most significant feature of EMT ( 24 26 ).…”

“…The abnormal expression of KIAA1199 has also been found in other cancer studies, such as oral squamous cell carcinoma ( Chanthammachat et al, 2013 ), breast cancer ( Evensen et al, 2013 ), gastric cancer ( Matsuzaki et al, 2009 ), colorectal tumors ( Tiwari et al, 2013 ; Birkenkamp-Demtroder et al, 2011 ; LaPointe et al, 2012 ), prostate cancer ( Michishita et al, 2006 ), ovarian cancer ( Shen et al, 2019 ) and hepatocellular carcinoma ( Jiang et al, 2018 ). It was reported ( Jiao et al, 2019 ) that KIAA1199 was abnormaly increased in the papillary thyroid tumor compared with normal specimens tissues and that upregulation of KIAA1199 was positively correlated with more advanced clinical variables.…”

“…Over-expression of KIAA1199 contributes to resistance to cell immortalization and cancerization in normal human cells and is associated with cell death ( Michishita et al, 2006 ). Several researches have illuminated that KIAA1199 is over-expressed in different cancers, including oral squamous cell carcinoma ( Chanthammachat et al, 2013 ), breast cancer ( Evensen et al, 2013 ), gastric cancer ( Matsuzaki et al, 2009 ), colorectal tumors ( Birkenkamp-Demtroder et al, 2011 ; LaPointe et al, 2012 ), prostate cancer ( Michishita et al, 2006 ), ovarian cancer ( Shen et al, 2019 ) and hepatocellular carcinoma ( Gu et al, 2018 ; Jiang et al, 2018 ). These studies showed that KIAA1199 regulates the proliferation, migration, and invasion of colorectal tumors, prostate cancer, ovarian cancer and so on.…”

“…CEMIP has been linked to the pathogenesis of a variety of malignancies in previous studies (9)(10)(11)(12)(13)(14); however, its role and mechanism in osteosarcoma remain unknown. To explore the effect of CEMIP on osteosarcoma, we detected the CEMIP expression in six pairs of osteosarcoma tissues and adjacent non-tumor tissues using qPCR and western blot, and found the CEMIP expression was significantly higher in the osteosarcoma tissues compared to the adjacent non-tumor tissues (Figures 1A-C).…”
