Key elements of the BMP/SMAD pathway co‐localize with CDX2 in intestinal metaplasia and regulate CDX2 expression in human gastric cell lines

Abstract: Helicobacter pylori infection induces intestinal metaplasia of the stomach, a preneoplastic lesion associated with an increased risk for gastric cancer development. Intestinal metaplasia is induced by the intestine-specific transcription factor CDX2 but the mechanisms responsible for this ectopic expression have never been described. We hypothesized that the BMP/SMAD pathway has a role in CDX2 regulation, in this context, for the following reasons: (1) the BMP pathway is crucial for normal intestinal different… Show more

“…CDX2 regulates CDH17 expression in colorectal cancer cell lines and gastric adenocarcinoma [Hinoi et al, 2002;Ko et al, 2005;Barros et al, 2008;Ge et al, 2008]. In this study, three putative CDX2-binding sites were found in the 1-kb CDH17 promoter region (Fig.…”

“…Recently, caudal-related homeobox 2 (CDX2) was found to play important roles in up-regulation of CDH17 expression in colorectal cancer cell lines and gastric adenocarcinoma [Hinoi et al, 2002;Ko et al, 2005;Ge et al, 2008], which was in turn regulated by bone morphogenetic protein 2 (BMP2) and BMP4 in gastric cancer cells [Barros et al, 2008]. Hepatic nuclear factor 1 (HNF1) is one of the key transcriptional factors (TF) that regulate serum a-fetoprotein levels in hepatocellular carcinoma [Nakabayashi et al, 2004].…”

“…24,25 As NKX6.3 did not affect BMP4 expression, we examined the effects of NKX6.3 on TWSG1, which is a novel inhibitor for the BMP signaling pathway 26,27 in AGS and MKN1 cells. Treatment with BMP4 induced p-SMAD1/5/8 and CDX2 expression and downregulated SOX2 expression in AGS and MKN1 cells (Figure 8a).…”

“…Eighteen hours posttransfection, cells were incubated in serum-free, Ca 2ϩ -free DMEM containing 4 mM L-glutamine, 0.2% BSA, penicillin 100 U/ml, and 0.5 mM CaCl 2 for 8 h. This medium was removed and substituted with the same medium supplemented with 0. 5 . The PCR reaction was prepared in a volume of 50 l containing the following reagents: 0.2 mM dNTP mixture; 1.5 mM MgCl2; 1 l template cDNA; 1.0 U platinum Taq DNA polymerase (Invitrogen, Carlsbad, CA).…”

“…CDX2 stimulates differentiation and expression of sucrase-isomaltase (SI), lactase-phlorizin hydrolase, LI-cadherin, and Muc2 in intestinal epithelial cells (12,31,71,79). BMP will increase CDX2 transcripts and protein in gastric cell lines (5). CDX2 expression is retained in the intestinal epithelium throughout adulthood but becomes reduced in colon cancer (21,27,52,71).…”
