Keratinization of the outer surface of the avian scutate scale: Interrelationship of alpha and beta keratin filaments in a cornifying tissue

Abstract: The outer surface of adult Gallus domesticus scutate scale was studied as a model for epidermal cornification involving accumulation of both alpha and beta keratins. Electron-microscopic analysis demonstrated that the basal cells of the adult epidermis contained abundant lipid droplets and that filament bundles and desmosomes were distributed throughout the cell layers. Indirect immunofluorescence microscopy and double-labeling immunogold-electron microscopy confirmed that the stratum germinativum contained al… Show more

“…The ultrastructural immunogold study confirms that there is a precise regulation of keratin synthesis from clear cells, which contain alphakeratin, and oberhautchen cells, where beta-keratin deposition starts. This sequence is different from that in crocodilians and birds, where beta-keratin is deposited over alpha-keratins from high suprabasal layers to prekeratinized layers where alpha-keratin almost disappears Shames et al, 1988Shames et al, , 1989Alibardi and Sawyer, 2002).…”

“…β-keratins have no molecular homology with α-keratins. They have a small molecular weight of about 10-25 kD and exhibit unique arrangements of pleated sheets (Shames et al, 1989;Presland et al, 1989a, b).…”

“…During formation of overlapping scutate scales the outer surfaces of these structures develop thick beta strata which accumulate polypeptides pl-7 (OGuin and O'Guin et al, 1982;Shames et al, 1988Shames et al, , 1989.…”
