Abstract Background Metastasis is the leading cause of melanoma mortality. Current therapies are rarely curative for metastatic melanoma, revealing the urgent need to identify more effective preventive and therapeutic targets. This study aimed to screen for the key core genes and molecular mechanisms related to the metastasis of melanoma. Methods Gene expression profile, GSE8401 including 31 primary melanoma and 52 metastatic melanoma clinical samples, was downloaded from the Gene Expression Omnibus (GEO). Differentially expressed genes (DEGs) between metastatic melanoma and primary melanoma were screened using GEO2R. Assays of gene ontology (GO), Kyoto Encyclopedia of Gene and Genome (KEGG) pathway and protein-protein interaction (PPI) were performed to visualize these DEGs through Database for Annotation, Visualization and Integrated Discovery (DAVID) software and Search Tool for the Retrieval of Interacting Genes (STRING) and Cytoscape with Molecular Complex Detection (MCODE) plug-in tools. Top 10 genes with high degree were defined as hub genes. Furthermore, paired post-metastatic melanoma cells and pre-metastatic melanoma cells were established by experimental mouse model of melanoma metastasis to verify the expression of these hub genes. Results 424 DEGs between the metastatic melanoma and primary melanoma were screened, including 60 upregulated genes enriched in ECM-receptor interaction and progesterone-mediated oocyte maturation and 364 downregulated genes enriched in amoebiasis, melanogenesis, and ECM-receptor interaction. CDH1, EGFR, KRT5, COL17A1, KRT14, IVL, DSP, DSG1, FLG and CDK1 were defined as the hub genes. . In addition, paired post-metastatic melanoma cells (A375M) and pre-metastatic melanoma cells (A375) were established and qRT-PCR analysis confirmed the expression of the hub genes during melanoma metastasis. Conclusion This bioinformatic study has provided a deeper understanding of the molecular mechanisms of melanoma metastasis. KRT5, IVL and COL17A1 have emerged as possible biomarkers and therapeutic targets in metastasis of melanoma.

CAF-1 è una proteina istonica trimerica implicata sia nella replicazione che nella riparazione del DNA, con il compito essenziale di stabilizzare la cromatina durante la replicazione; presenta un azione di assemblaggio del tutto tipica, poiché unisce solo DNA che è andato incontro a replicazione. Di recente la proteina CAF-1p60 è stata proposta come marker della proliferazione cellulare nei tumori solidi, in particolare nel distretto testa- collo in virtù della sua iper-espressione nelle cellule in stato di proliferazione rispetto a quelle quiescenti in cui è down- regolata. Questa relazione con lattività mitotica ha inoltre permesso di considerarla come possibile indice prognostico di aggressività neoplastica. In precedenti lavori, effettuati presso il nostro dipartimento, abbiamo documentato che tale proteina risulta essere iper-espressa nei tumori del cavo orale, delle ghiandole salivari e della tiroide. In questo studio abbiamo analizzato e confrontato lespressività immunoistochimica della proteina CAF-1/p60 nelle neoformazioni laringee precancerose, nei carcinomi in situ e nei tumori maligni, in particolare: in 30 casi di displasia moderata e/o severa, 30 casi di carcinoma in situ e 30 casi di SCCs. CAF-1/p60 è iperespressa nelle cellule neoplastiche; lespressione di CAF-1/p60 aumenta significativamente in correlazione allalto indice di replicazione cellulare; inoltre la sua iperespressione moderata-severa è correlata con una prognosi peggiore comparata con la lieve e liperespressione della proteina CAF-1/p60 è correlata con un più alto rischio di morbilità e mortalità rappresentando un reale indice prognostico delle neoplasie laringee. CAF-1/p60 assume, pertanto, un valore prognostico indipendente nelle neoplasie laringe. Riteniamo, dunque, che liperespressione della proteina CAF-1/p60 che possa essere usata come indicatore di aggressività nellevoluzione maligna specie dei carcinomi in situ ed impiegata nel follow-up per identificare le forme a più alto rischio prognostico che necessitano quindi controlli più ravvicinati nel tempo.

Background: Squamous metaplasia (SM) is an irreversible form of airway epithelial remodeling. Hyperproliferation of basal cells was observed in squamous metaplastic epithelium of chronically inflamed airway. However, the association of such aberrant proliferation of basal cells with SM in the nasal epithelium after radiation damage remains unclear. The aim of this study was to investigate SM and accompanying levels of p63+Krt5+ (basal cell markers) cells in the nasal epithelium of patients with radiation-induced chronic rhinosinusitis (CRSr) and patients with chronic rhinosinusitis without nasal polyps (CRSsNP) compared to healthy controls.Methods: We assessed the prevalence of SM and the expression of p63+, Krt5+, p63+Krt5+, and Ki67+ cells through immunofluorescence(IF) staining of the inferior turbinate (IT) tissues from patients with CRSr (n = 36) , CRSsNP (n = 33) and controls (n = 28).Results: The prevalence of SM and the number of p63+Krt5+ cells were both significantly increased in patients with CRSr compared to patients with CRSsNP and controls. The number of Ki67+ cells were both significantly increased in patients with CRSr and CRSsNP compared to controls, but the ratio of Ki67+ cells to p63+Krt5+ cells was significantly lower in patients with CRSr compared to patients with CRSsNP. In patients with CRSr, an increased number of p63+Krt5+ basal cells was observed in SM epithelium compared to non-SM epithelium.Conclusion: SM is increased in the nasal epithelium of patients with CRSr, in which aberrant levels of p63+Krt5+ basal cells serves as an important pathologic feature in the squamous metaplastic epithelium.
