Jian-Pi-Yi-Shen Formula Regulates Inflammatory Cytokines Production in 5/6 Nephrectomized Rats via Suppression of NF-κB Activation

Abstract: Jian-Pi-Yi-Shen formula (JPYSF) is a Chinese herbal decoction used for treating chronic kidney disease (CKD) for over 20 years with good efficiency. However, the mechanism lacks solid evidence. In the present study, we tested the hypothesis that JPYSF may retard CKD progression via inhibition of inflammation in 5/6 nephrectomy (5/6 Nx) rat model. The 5/6 Nx rats were randomly divided into 2 groups: 5/6 Nx group and JPYSF group. Sham-operated rats served as control. JPYSF (2.06 g/kg/d) were administrated by gav… Show more

“…These effects could be involved with the production of EPO via HIF signaling pathway. In line with these findings, our previous studies revealed that JPYS remarkably retards development and progression of CKD in animal model and induced expression of EPO in cultured kidney cells [9, 10, 12]. The results of this study demonstrated that the serum EPO levels were decreased in CKD rats.…”

“…Besides, the overexpression of serum hepcidin reverted to its original level by JPYS treatment, ensuring the systemic and intracellular iron homeostasis. In line with this notion, previous studies showed that JPYS could suppress proinflammatory cytokines production in 5/6 nephrectomized rats [ 36 ]. The hepcidin expression in the liver can be induced by the upregulation of cytokines, most notably IL-6 and LPS [ 27 ], whereas the increased systemic hepcidin level can act on macrophages, preventing the release of iron recovered from senescent red blood cells into plasma [ 37 ].…”

“…Sections were stained with periodic acid–Schiff (PAS) and Masson’s trichrome stains. The degree of tubular atrophy in PAS staining was determined based on a scale of 0–3 points: 0, normal tubules; 1, rare single atrophic tubule; 2, several clusters of atrophic tubules; and 3, massive atrophy (Lu et al, 2018). Interstitial fibrosis area in Masson staining was measured using Image J software (NIH, Bethesda, MD, USA).…”
