Jasmonoyl-l-isoleucine is required for the production of a flavonoid phytoalexin but not diterpenoid phytoalexins in ultraviolet-irradiated rice leaves

Abstract: Rice produces low-molecular-weight antimicrobial compounds known as phytoalexins, in response to not only pathogen attack but also abiotic stresses including ultraviolet (UV) irradiation. Rice phytoalexins are composed of diterpenoids and a flavonoid. Recent studies have indicated that endogenous jasmonyl-l-isoleucine (JA-Ile) is not necessarily required for the production of diterpenoid phytoalexins in blast-infected or CuCl2-treated rice leaves. However, JA-Ile is required for the accumulation of the flavono… Show more

“…Root samples were harvested 1–12 mm from the root–shoot junction at 0, 2 and 8 h after root cutting, and frozen in liquid nitrogen. The hormone analysis was carried out as described previously ( Miyamoto et al 2016 , Enomoto et al 2017 ). Briefly, samples of approximately 100 mg FW were suspended in 80% (v/v) aqueous methanol with [ 13 C 6 ]IAA, [ 2 H 2 ]JA and [ 13 C 6 ]JA-Ile as internal standards.…”

“…The elicitation was considered to arise from the induction of naringenin 7- O -methyltransferase, a key enzyme in sakuranetin biosynthesis [156]. Very interestingly, JA-Ile was not required for diterpenoid production in blast pathogen-infected or CuCl 2 -treated rice leaves [157].…”

“…In the final step of sakuranetin biosynthesis, the precursor naringenin is converted to sakuranetin by naringenin 7- O -methyltransferase (NOMT), which is encoded by OsNOMT 11. We recently reported that OsNOMT expression is induced by JA treatment12, and jasmonoyl- l -isoleucine (JA-Ile), an active form of jasmonate, is required for the production of sakuranetin10. We have previously shown that CuCl 2 is a potent elicitor to induce sakuranetin production in wild-type plants, while such CuCl 2 -inductive production of sakuranetin is specifically compromised in a JA-Ile deficient mutant osjar1 10.…”
