Ivabradine protects rats against myocardial infarction through reinforcing autophagy via inhibiting PI3K/AKT/mTOR/p70S6K pathway

Abstract: Ivabradine (Iva), a heart rate reducing agent that specifically inhibits the pacemaker I(f) ionic current, has been demonstrated to be cardioprotective in many cardiovascular diseases. Autophagy is an evolutionarily conserved metabolic process that regulates cardiac homeostasis. This study is aimed to explore whether autophagy is functionally involved in the cardioprotective effect of Iva in a rat model of myocardial infarction (MI). We observed that Iva treatment (po, 10 mg/kg/day) showed significant recovery… Show more

“…Akt serves as a downstream serine/threonine kinase of PI3k and involves in cell proliferation and differentiation by activating downstream factors HIF-1a and mTOR [ 31 , 32 ] . PI3K/AKT works as an important signal regulatory mechanism for human cell growth and development [ 33 , 34 , 35 ] . It mediates diverse cellular processes including cell growth, development, division, differentiation, apoptosis, and even intercellular functions.…”

“…On the contrary, Ivabradine treatment reversed these changes. Consistently, the activated levels of PI3K, AKT, mTOR and p70S6K were increased in the myocardial infarction -subjected rat heart tissue but Ivabradine treatment inhibited these changes, thus enhancing autophagy ( Dai et al, 2021 ). A similar effect was seen in a study by Yu et al, who investigated Ivabradine in the cardiac model of TAC.…”

“…AKT/mTOR is an essential signaling pathway for regulating cellular glucose metabolism and maintaining cell growth and reproduction [46], and is a recognized oncogenic driver [47] which mediates metabolic imbalance and regulation of the cell cycle to bene t the survival and growth of cancer cells [48,49]. The activation of AKT/mTOR signaling pathway plays a role in inhibiting apoptosis and protecting cardiomyocytes [50]. However, there is insu cient evidence to describe its relationship with ZYX and its role in HCC.…”
