iTRAQ‐Based Proteomic Analysis Exploring the Influence of Hypoxia on the Proteome of Dental Pulp Stem Cells under 3D Culture

Abstract: Hypoxic preconditioning is commonly applied to enhance mesenchymal stem cells (MSCs) therapeutic effect before transplantation. Elucidating the effect of hypoxic preconditioning would be beneficial for improved application. However, the influence of hypoxia on dental tissue derived MSCs cultured in 3D was unknown. Thus, the present study is to investigate gene expression and proteome of dental pulp stem cells (DPSCs) after hypoxic preconditioning. DPSCs were isolated, cultured in a 3D system under the normoxic… Show more

“…It has been used to induce chemical hypoxia for studying stem cells. It has demonstrated responses similar to those produced by cells subjected to hypoxia [ 21 , 32 ]. The mechanism of action that CoCl 2 uses in producing a hypoxic environment is attributed to the substitution of iron by cobalt ions in prolyl hydroxylases (PHDs).…”

“…When passaging the cells, a mixture of 0.25% trypsin and 0.01% EDTA was used. The expression of CD markers in these HDPCs was analyzed and published in a previous study by our group (7). For all experiments, the HDPCs were in passages 3-6.…”

“…Dou et al[ 37 ] deliberately examined the impact of momentary hypoxia on the gene expression and proteomic profile of human DPSCs under 3D culture conditions. Most of the upregulated proteins engaged in cell processes, angiogenesis, protein binding and transport, direction of reaction to stimuli, metabolic processes, and immune responses.…”
