The sweet melon fruit is characterized by a metabolic transition during its development that leads to extensive accumulation of the disaccharide sucrose in the mature fruit. While the biochemistry of the sugar metabolism pathway of the cucurbits has been well studied, a comprehensive analysis of the pathway at the transcriptional level allows for a global genomic view of sugar metabolism during fruit sink development. We identified 42 genes encoding the enzymatic reactions of the sugar metabolism pathway in melon. The expression pattern of the 42 genes during fruit development of the sweet melon cv Dulce was determined from a deep sequencing analysis performed by 454 pyrosequencing technology, comprising over 350,000 transcripts from four stages of developing melon fruit flesh, allowing for digital expression of the complete metabolic pathway. The results shed light on the transcriptional control of sugar metabolism in the developing sweet melon fruit, particularly the metabolic transition to sucrose accumulation, and point to a concerted metabolic transition that occurs during fruit development.

Hexokinase (HXK, EC 2.7.1.1) plays an important role in the metabolism and glucose signalling. To examine the characteristics of HXK gene family in rice, the subcellular localizations of ten hexokinases (OsHXK1 -OsHXK10) were determined using OsHXK::GFP fusion proteins in tobacco mesophyll protoplasts. As was previously demonstrated, OsHXK4 was detected in the chloroplast stroma, OsHXK5 and OsHXK6 in the mitochondria, and OsHXK7 and OsHXK10 in the cytoplasm. In the present study, OsHXKs were clearly divided into three types (A, B, C) based on their N-terminal sequences. The new type-C HXKs in plants, OsHXK1, OsHXK7 and OsHXK8, which lack the plastidic transit peptide and the membrane anchor domain, were detected not only in the cytoplasm but also in the nucleus. The type-B HXKs, OsHXK2, OsHXK3, OsHXK9 and OsHXK10, which contained a membrane anchor domain, were distinctly localized in the mitochondria. These results suggest that OsHXKs localized in different cell compartments may be involved in the glucose signalling-related gene expression during growth and development of rice.

The use of sugarcane as a biofactory and source of renewable biomass is being investigated increasingly due to its vigorous growth and ability to fix a large amount of carbon dioxide compared to other crops. The high biomass resulting from sugarcane production (up to 80 t/ha) makes it a candidate for genetic manipulation to increase the production of other sugars found in this research that are of commercial interest. Sucrose is the major sugar measured in sugarcane with hexoses glucose and fructose present in lower concentrations; sucrose can make up to 60% of the total dry weight of the culm. Species related to modern sugarcane cultivars were examined for the presence of sugars other than glucose, fructose and sucrose with the potential of this crop as a biofactory in mind. The species examined form part of the Saccharum complex, a closelyrelated interbreeding group. Extracts of the immature and mature internodes of six different species and a hybrid were analysed with gas chromatography mass spectrometry to identify mono-, di-and tri-saccharides, as well as sugar acids and sugar alcohols. Thirty two sugars were detected, 16 of which have previously not been identified in sugarcane. Apart from glucose, fructose and sucrose the abundance of sugars in all plants was low but the research demonstrated the presence of sugar pathways that could be manipulated. Since species from the Saccharum complex can be interbred, any genes leading to the production of sugars of interest could be introgressed into commercial Saccharum species or manipulated through genetic engineering.
