“…Different authors have shown the interest of enzyme electrophoresis for the studies of cassava genetics, although some phenotypes were unstable depending on the origin of the extract and the buffers (Zoundjihekpon & Tour6, 1983;Ramirez et al, 1987). In our study, we were able to give a genetic interpretation for most of the phenotypes, after fixing standard conditions for extraction, migration and staining.…”

“…The first attempts to use enzyme electrophoresis on cassava showed a high level of polymorphism within the species (Zoundjihekpon & Tour6, 1985;Hussain et al, 1987;Ramirez et al, 1987). We used starch gel electrophoresis as a cheap and simple technique to identify genetic markers from 10 enzyme systems.…”

“…Agronomical and botanical descriptors (Pereira et al, 1992;Cury, 1993) and isoenzyme analysis (Ramirez et al, 1987;Léfèvre et al, 1993) have been developed to evaluate genetic diversity. However, a limited number of isoenzymatic systems (Pasteur et al, 1987) and botanical descriptors (Cury, 1993) are available; therefore, polymorphisms are restricted.…”

“…For the molecular characterization, we used seeds and seedlings of 443 individuals coming from six populations collected in 1987, 1998, Zarcero (48), Aserrí (84), Tarbaca (18), Jérico (64), and Quircot (119). Seed storage protein phaseolin and two enzymes (diaphorase and peroxydase) were analyzed following the protocol by Lareo et al (1993) and Ramírez et al (1987), respectively. The interpretation of the diaphorase pattern was done along Sprecher (1988), while we followed the procedure by Koenig & Gepts (1989) for peroxydase.…”
