Isotypic Analysis of Grass-Pollen-Specific Immunoglobulins in Human Plasma

Abstract: Previously, the specificity of human immune responses to Dactylis pollen was analyzed in 26 plasma samples with high levels of grass-pollen-specific IgG4 (‘IgG4+ plasma’, largely from grass-pollen-allergic patients), as compared to 25 plasma samples with low grass-pollen-specific IgG4 (‘normal plasma’, from nonatopic individuals). In the present study, a quantification of the Dactylis-pollen-specific IgE, IgM, IgA class and IgG subclass antibodies in these plasma samples is proposed. Isotypic distribution in I… Show more

“…The earliest recognition of IgG4 in allergies was from Allergen-specific immunotherapy (AIT) data, demonstrating a tolerance-inducing function (14)(15)(16). However, later clinical observations revealed that allergen-specific IgG4 was associated with allergic sensitization and correlated to allergic diseases, such as allergic rhinitis (AR), asthma, atopic dermatitis (AD), and anaphylaxis (17)(18)(19)(20)(21)(22)(23). IgG4 levels may vary significantly in healthy individuals (24), strongly hindering its clinical application as a diagnostic tool.…”

“…Aside from the clinical implications, aeroallergens are non-infectious environmental antigens to which most people will mount a minimal T-cell-dependent and allergen-specific humoral response involving the IgA and IgG isotype classes [5, 6]. We have shown that atopic and non-atopic humans can mount equally vigorous humoral responses to these challenges [7, 8].…”
